Trying to animate this tentacle but only the first bone seems to be doing anything. What did I do wrong?

It looks like only the first bone is "rested". You need to select all bones in the Scene Editor, press "r" to set their rest position. I'd set the rotation on the first bone back to 0 (from 90 where it's set in the scene) before doing so, though.

Attached is a modified version of your scene with 1) first bone set to 0 degrees heading, 2) all bones rested, 3) all bones set to Inverse Kinematics in their Motion Options=>Controllers and Limits panel, 4) a NULL named Goal added, and 5) Bone.000(15) set to have Goal as it's IK Goal.

When you move the Goal null around, the tentacle will follow it. Obviously, if you want to manually animate the bones you can either avoid IK altogether, or use the IK/FK mix to do a bit of both.
